De novo transcriptome sequencing and anthocyanin metabolite analysis reveals leaf color of Acer pseudosieboldianum in autumn

Abstract Background Leaf color is an important ornamental trait of colored-leaf plants. The change leaf closely related to the synthesis and accumulation anthocyanins in leaves. Acer pseudosieboldianum a tree native Northeastern China, however, there was less knowledge about biosynthesis many steps pathway remain unknown date. Results Anthocyanins metabolite transcript profiling were conducted using HPLC ESI-MS/MS system high-throughput RNA sequencing respectively. results demonstrated that five detected this experiment. It worth mentioning Peonidin O-hexoside Cyanidin 3, 5-O-diglucoside abundant, especially displayed significant differences content at two periods, meaning it may be play role for final color. Transcriptome identification showed total 67.47 Gb clean data obtained from our results. Functional annotation unigenes, including comparison with COG GO databases, yielded 35,316 unigene annotations. 16,521 differentially expressed genes identified statistical analysis gene expression. formation PAL, ANS, DFR, F3H selected. Also, we screened out regulatory such as MYB, bHLH WD40. Combined detection metabolites, pathways anthocyanin analyzed.
